FDA Relaxes Hold on Filana’s Simufilam TSC Trial

The U.S. Food and Drug Administration (FDA) has lifted a clinical hold on Filana Therapeutics Inc.’s Phase 2a trial of simufilam, an oral small-molecule candidate designed to treat epilepsy in patients with tuberous sclerosis complex (TSC). The trial, conducted at 13 U.S. sites, will evaluate two doses of simufilam in 40 subjects aged 12 to 55 years, focusing on seizure frequency, intensity, and related outcomes. Participants who complete the 16-week double-blind phase may enter a 48-week extension study. TSC, a rare genetic disorder affecting around 45,000 Americans, is linked to mutations in the TSC1 or TSC2 genes, with 80% to 90% of patients experiencing epilepsy. Despite multiple approved treatments, more than 60% of TSC patients remain refractory to antiepileptic therapy. Preclinical studies in mouse models demonstrated simufilam’s potential to reduce seizure frequency and progression in TSC-related pathology. Filana holds an exclusive worldwide license from Yale University for the drug’s development in TSC-related epilepsy.
