Syndax Pharmaceuticals presented at Citigroup’s Biopharma Back to School Summit on Thursday, highlighting strong commercial traction for its menin inhibitor revumenib, known commercially as Revuforj.

Chief Executive Officer Michael Metzger said the drug is annualizing at “well north of $200 million” in its second year since launch, having generated $55 million in the latest quarter. The company holds roughly 85% market share across the KMT2A and NPM1 indications in relapse-refractory acute myeloid leukemia — far outpacing a competitor in the same space that posted about $9 million in the comparable period.

“Thinking about relapse-refractory, I don’t think you can get more dominant than that,” Metzger said.

Syndax projects a U.S. peak revenue opportunity for Revuforj exceeding $2 billion if the therapy reaches both fit and unfit patient populations in the relapse-refractory setting. About 50% of Revuforj-treated patients proceed to transplant, and the company aims for maintenance restart rates of 70% to 80% post-transplant. Patients on post-transplant maintenance are averaging more than nine months on therapy, while duration in the relapse-refractory indication is expected to land in the six- to 12-month range.

Also discussed was Revuforj’s potential in patients carrying NUP98 mutations, which may account for 3% to 5% of AML cases. Data presented at the European Hematology Association showed a little more than a 25% response rate in a 28-patient dataset.

On the pipeline front, two newer programs — SNDX-4321 targeting lung cancer and SNDX-62122 for myelofibrosis — are expected to enter the clinic in 2027, according to management.

Revumenib is also being evaluated in IPF through the MaxBio program. A Phase II proof-of-concept trial enrolled approximately 135 patients in a randomized, 2-to-1 placebo-controlled, double-blind design, over-enrolled by roughly 10 patients. Management indicated that showing roughly 30 to 40 milliliters of absolute forced vital capacity retention — or about 40% relative retention — would be an encouraging result in the IPF indication.

Chief Medical Officer and Head of R&D Nick Botwood called the IPF data “a novel and groundbreaking outcome for a completely new modality” and described the EGFR lung cancer program as “an incredible opportunity to validate an entirely new hypothesis.”

Shares of Syndax closed at $20.56, down $0.12 or 0.58%, according to InvestingPro data. Wall Street price targets range from $26 to $57.
