Rani Therapeutics, a company developing oral biologics, presented its progress at the H.C. Wainwright 28th Annual Global Investment Conference. The company's RaniPill technology, which delivers monoclonal antibodies and large proteins through a swallowable auto-injector, has shown promising results in clinical trials.

Shares of Rani Therapeutics were trading at $0.84, with a market capitalization of approximately $117 million. The company ended the second quarter with $53 million in cash, providing a financial runway through the end of 2027. Earlier in 2024, Rani Therapeutics raised $20 million, followed by a $60 million PIPE financing in late 2024, backed by investors like Samsara and RA Capital.

The company has a multi-target collaboration with Chugai, potentially worth $1 billion, which includes one rare-disease license and five option agreements for high-value immunology targets. The stock has delivered a 69% return over the past year, with Wall Street analyst price targets ranging from $5 to $11 per share. Revenue growth has been noted at over 300% in the last twelve months.

Rani Therapeutics' lead program, RT-114, showed more than 150% relative bioavailability compared to subcutaneous dosing in Phase I-A data. Adverse events for RT-114 were comparable to or lower than a matched subcutaneous dose. A 24 mg expansion cohort was added in Phase I-B to study dose response, peak concentration, and tolerability, with efficacy data expected in the first half of 2025.

The company's Fremont, California facility, the first fully automated manufacturing line designed for a capacity of 1,500 pills a day, is intended to support all rare-disease programs and Phase III studies. Future planned capacity is targeted at 10,000 pills a day for peak immunology commercialization, with deployment and qualification completion expected by the end of 2027.

CEO Talat Imran highlighted the RaniPill's mechanism, stating it looks like a pill but is essentially a swallowable auto-injector. He noted that Chugai conducted extensive due diligence, testing two of their molecules in the RaniPill and reviewing the company's manufacturing and quality processes. Imran also emphasized the potential of RT-114, stating the results were surprising, showing greater than 150% relative bioavailability compared to subcutaneous dosing.

Imran also discussed the company's weight-loss positioning, noting that Rani Therapeutics is not aiming to compete directly with tirzepatide or Wegovy but aims to offer a competitive product with an attractive adverse event profile.
